In May 2021, Microread Genetics signed an innovative cooperation agreement with Dubai police G.H.Q. The two parties will carry out in-depth cooperation in the field of forensic science based on their respective advantages and jointly develop a rapid mutation Y chromosome detection STR kit, to improve the efficiency of forensic case detection and family investigation in the Middle East and the whole world.

After nearly a year of joint efforts, the cooperative kit has been successfully developed and transferred to production, officially named "Microreader™ 26 RM-Yplex ID system". On the WPS2022, Mrs. Hanan A. Almulla, a technical expert at the Forensic Training and Development Center of Dubai Public Security Headquarters, delivered a wonderful report entitled "Development of novel single multiplex system incorporating 26 rapidly mutating Y STRs" on behalf of Dubai police G.H.Q.

 

In the report, Mrs. Hanan A. Almulla first introduced the wide application of Y-STR in forensic science. At present, the Y chromosome STR kit is mainly used in case scenarios such as family distinction and suspected family location, especially in the detection of sexual assault cases, when high female DNA noise exists, it can play a very good typing role. However, the Y-STR kit also has its limitations in practical application. For example, male suspects in a family usually have completely consistent Y-STR types, and conventional Y-STR loci are difficult to distinguish different types in the same family. male individual. Rapidly mutating Y-STRs [Rapidly mutating (RM) Y-STRs, RM-Y] can be used to distinguish male related individuals in the same family because of their higher polymorphism. In the past ten years, scientists from many countries have devoted themselves to finding a better RM-Y STR and trying to construct a multi-site multiplex amplification system in order to significantly increase the ability to identify male families. But so far there is still no single-tube multiplex amplification system with a large number of RM-Y sites.

With the efforts of Dr. Rashed Alghafri of Dubai Police G.H.Q and the R&D department of Microread genetics, the two parties jointly developed a mature commercial kit that can amplify 26 rapidly mutating Y-STR sites in a single tube, and strictly followed the international SWGDAM guidelines. Key performances such as sensitivity, stability, repeatability, mixed sample detection rate and species specificity were verified. The results show that the Microreader™ 26 RM-Yplex ID system can still obtain complete typing when the DNA amount is as low as 0.125ng, and it can be used in mixed samples of male and female at a ratio of 1:3200, and samples of mixed male and female at a ratio of 1:9 , successfully obtained the complete typing result. At the same time, the kit has excellent anti-inhibitor ability.

 

This kit is a powerful weapon for distinguishing different male related individuals in the same family in cases, especially sexual assault cases. Both Microread Genetics and the Dubai Police G.H.Q hope that this kit can be widely used around the world to help public security organs solve more complex and difficult cases.
